Steroidogenic Acute Regulatory Protein (STAR) controls the rate-limiting step of steroidegenesis by translocating cholesterol from the outer mitochondrial membrane to the inner membrane where it is later cleaved to pregnenolone. It is primarily present in steroid-producing cells, including Leydig cells in the testis, theca cells and luteal cells in the ovary and adrenal cells in the adrenal cortex. Due to low levels of pregnenolone, seminomas and Leydig cell tumors display no specific STAR staining. Therefore, STAR antibody may assist in differentiating sex cord stromal tumors (SCST), seminomas and embryonal carcinomas. This antibody recognizes a protein of 83 kDa, which is identified as Factor XIIIa. It has been identified in platelets, megakaryocytes, and fibroblast-like mesenchymal or histiocytic cells in the placenta, uterus, and prostate, monocytes and macrophages and dermal dendritic cells. Anti-factor XIIIa has been found to be useful in differentiating between dermatofibroma (almost all cases are positive), dermatofibrosarcoma protuberans (-/ ) and desmoplastic malignant melanoma (-). The major constituents of mucus, the viscous secretion that covers epithelial surfaces such as those in the trachea, colon, and cervix, are highly glycosylated proteins called mucins. These glycoproteins play important roles in the protection of the epithelial cells and have been implicated in epithelial renewal and differentiation. This gene encodes an integral membrane glycoprotein found on the cell surface, although secreted isoforms may exist. MUC-4 transcripts have been detected in normal respiratory epithelium and lung. MUC-4 is a very specific (100%) and sensitive (90%) marker of lung adenocarcinomas and is negative for mesotheliomas. Reportedly, MUC-4 expression in invasive ductal carcinoma of the pancreas is an independent factor for poor prognosis. CHD4/Mi-2b is a component of the nucleosome remodeling and deacetylase (NuRD) complex which is a multi-subunit protein complex containing both histone deacetylase and nucleosome-dependent ATPase subunits. Current models predict that this complex function primarily in transcriptional repression. Accumulating evidence indicates that NuRD may regulate the transcription of specific genes by interacting with specific transcriptional factors. In addition, the NuRD complex may also participate in genome-wide transcriptional regulation through an association with histone tails.
